蛋白酶是生物系統(tǒng)生長(zhǎng)發(fā)育、營養(yǎng)代謝等活動(dòng)中必不可少的基礎(chǔ)大分子催化物質(zhì),是生命醫(yī)學(xué)領(lǐng)域一直以來關(guān)注的重點(diǎn)。研究表明,大多數(shù)疾病的發(fā)生伴隨著蛋白酶的表達(dá)異常,某些特定蛋白酶成為一些疾病的生理標(biāo)志物,因此開發(fā)蛋白酶響應(yīng)診療探針可對(duì)相應(yīng)的疾病診斷和治療提供幫助。熒光成像具有實(shí)時(shí)、無創(chuàng)、空間分辨率高等特性,而納米診療探針通過合理設(shè)計(jì)可實(shí)現(xiàn)蛋白酶響應(yīng)熒光成像的同時(shí)整合其它治療方式于同一納米平臺(tái),達(dá)到成像和治療一體化的良好效果?偨Y(jié)了靶向基質(zhì)金屬蛋白酶、組織蛋白酶、半胱天冬氨酸蛋白酶、胰蛋白酶和一些其它類型(如成纖維細(xì)胞激活蛋白等)蛋白酶的納米熒光診療探針的成像設(shè)計(jì)原理及生物醫(yī)學(xué)應(yīng)用,希望促進(jìn)新型低毒、高性能的蛋白酶響應(yīng)型納米熒光探針的開發(fā),并早日實(shí)現(xiàn)臨床轉(zhuǎn)化。

Protease is an essential basic macromolecular catalyst for the growth and development of biological system and nutritional and metabolic activities. It has always been the focus of life medicine and other fields. Studies have shown that most diseases are accompanied by abnormal expression of proteases, so some specific proteases have become physiological markers of some diseases. The development of protease detection tools can help in the diagnosis and treatment of corresponding diseases. Fluorescence imaging has the characteristics of real-time, noninvasive and high spatial resolution. Through reasonable design, the fluorescent nano diagnosis and treatment probe can realize the response imaging of protease precise targeting. At the same time, based on nano structure, it can combine other treatment methods on the platform to achieve the good effect of imaging and treatment integration. In this review, we summarize the imaging design principles and biomedical applications of nano fluorescent diagnostic probes targeted to matrix metalloproteinases, cathepsins, caspases, trypsin and some other types of proteases such as fibroblast activating protein, hoping to promote the production of new protease responsive nano fluorescent probes with low toxicity and high performance, and realize clinical transformation as soon as possible.
